Your sequences, in bulk

Custom bulk ASO synthesis

Paste or upload a list of 5 or more sequences in any notation. The oligonucleotides are made HPLC-purified in the chemistry you choose, in tubes or on plates at the same price per oligo.

Paste your sequences in any notation, choose how they are made, read the list back, and send the request.

01 Your sequences

Species, for the names in the list (optional)

An example, not your list

a few sequences in the notations the reader accepts, to show how a list is read back.

From 5 to 384 sequences, each read by the reader the sequence builder uses: plain letters, slash codes, bracket codes, mA or +A prefixes, an asterisk or an s for a phosphorothioate linkage, 8 to 110 nucleotides. Typed, pasted, or from a file: one sequence per row, a name before a comma or a tab if you like. A file of up to 500 rows and 1 MB, as .txt, .csv or .tsv; a spreadsheet workbook is refused, so save it as CSV first.

A bulk list starts at 5 sequences. A shorter list, or one sequence only, is a single-sequence order. The order page prices a single set of sequences.

02 Chemistry

Plain letters take this chemistry; a row that names another is read as written and quoted.

03 Purification

HPLC, the purification every screening product is made in, in tubes and on plates alike: one grade for the whole screen.

04 Format

A delivery format at no difference in price: every oligo is priced the same in tubes and on a plate. A plate is available from 48 oligos, one per sequence; below that the order is delivered in tubes. A plate is filled down the columns with its control wells at the end.

5 oligos on this list: delivered in tubes, a plate being available from 48.

Plain letters in the 2'-MOE and locked nucleic acid chemistries are painted

05 Controls, included

In tubes, the class's scramble control alone, one per order, after the samples, at no charge, from the catalogue's own HPLC controls. The positive control and the labeled scramble travel with a plate; the order page sells them on their own.

Capabilities

ASO manufacturing capabilities

From research validation to preclinical development

AUM BioTech provides research-grade antisense oligonucleotides with chemical modification capabilities for research and preclinical development. Our scalable manufacturing platform delivers consistent quality from research scale (μmol) through multi-gram production, with validated analytical methods and complete analytical documentation.

Whether you are advancing a target from research validation to preclinical studies, or need larger quantities of a modified ASO for extended in vivo work, our custom synthesis service delivers the exact modifications, scale, and purity you need.

Modifications

Modification capabilities

A bulk order can combine sugar and backbone modifications position by position, from those below.

Sugar modifications

2'-O-methyl (2'-OMe)
Resists nucleases, and an oligonucleotide modified at every position does not support RNase H cleavage
2'-O-methoxyethyl (2'-MOE)
More nuclease resistance than 2'-OMe
2'-fluoro (2'-F)
Raises affinity for the target RNA
Locked nucleic acid
More affinity per residue than the other three

Backbone modifications

Phosphorothioate (PS)
Resists nucleases and binds proteins, lowers duplex affinity slightly, and can raise toxicity at high concentration
Phosphodiester (PO)
The unmodified backbone, digested quickly in serum
Mixed PS/PO
Phosphorothioate at the chosen positions only
Custom chimeric designs
Sugar and backbone set position by position

Scale

Flexible scale options

Scaling from research to preclinical production

Flexible scale options
Scale tierQuantity rangeTypical applicationsTurnaround
Research scale1-10 μmolTarget validation, initial screening, mechanism studies2-3 weeks
Pre-clinical10-100 μmolIn vivo efficacy, PK/PD studies, dose-response optimization3-4 weeks
Scale-up100-1000 μmolPreclinical toxicology, formulation development, scale-up material6-8 weeks
Bulk production1-500+ gramsExtended in vivo programs, multi-study preclinical campaigns, reference and stability material8-24 weeks

A bulk order follows the turnaround in this table, not the standard lead time of 10-14 business days the catalogue's products carry.

For a scale beyond the catalogue's ladder, write it in the notes of the request above.
